1980 with 68K miles, mostly/all stock

Weekend driver — aspire for 250mile road trip ability ...

What should be my regular maitenace routine?
- every xx miles or XX months have shop do?

What should I do before attempting longer haul / 250 mile road trip?
- besides obv asking mechanic if she’s good to go?

All depends. Do you hear any loud whining noises while at highway speeds? If you don't blow smoke, or loose water, transmission shifts well, go for it. Check all fluids before leaving, and carry spare fluids in case of emergency.
