I'd agree with everything said so far but I must highlight two things...first off is mileage, as said above it doesn't seem to change regardless of if you're pulling a trailer, unloaded, uphill, downhill, it just doesn't change...which is nice.
Secondly is the seat of your pants grunt all the way through the power band...our 2000 3/4 ton pickup has the vortec 454, 4L80E, 4.10's and like 32"ish tall tires and I'll tell you what...that truck will put you in your seat no matter where you're @ in the throttle...no replacement for displacement :)
 
My brotther's truck is pretty close to mine, both k5s , only a 200 lbs. diff on a scale and almost the same gear to tire ratio. mine is 42" with 4.88, his is 39" with 4.10s. the real diff is the motor. i have the o.e. 350 in mine with 265k miles on it. I tore it down and recammed it, put headers and exhaust and an intake on it. his has a 388" stroker with all the goodies. i can take him from stop light to stop light as i have no problem reving mine to 5800, but any where else like a rolling acceleration or up a long grade to the mountain, and he leaves me for dead. steep long uphills make mine struggle to maintain 65mph, my brother passes me and laughs because he still has is at less than half throttle.

I'm thinking 427 with a roller cam and all the goodies. I like the 427 for the mid and the top end over the 454. they also rev alot quicker and seem to go longer between rebuilds.
