you put the tailgate down, take off the access panel (there are about 12-18 sheet metal screws holding it down), roll up the window a few inches so you can get to the 7/16" bolts at the bottom channel, take those out and it should come out.

depending on how loose you window sweeps are you might have to remove 1 to get the channel past those.

Just sit backwards on your seat and pry up the corner of the gasket. As you roll it up, push it out of the cab. Should come out very easy (but might want to have someone outside to catch the glass).
 
either i didnt read pickup earlier or it was changed.
either way i just assumed it was a blazer, probaly cause thats what i have, oops. /forums/images/graemlins/crazy.gif
 
Being that it was edited I'd say you probably were replying at the same time he was changing it to Pickup /forums/images/graemlins/rotfl.gif
